The Plasmin Inhibitor of Human Plasma
نویسندگان
چکیده
Purified al-proteolytic inhibitor from human plasma was shown to inhibit trypsin, chymotrypsin, plasmin, and thrombin. This inhibitor is probably identical with the inhibitor previously known as serum trypsin inhibitor, al-antitrypsin, or antiplasmin. The enzymes were affected by the inhibitor via two different mechanisms. Trypsin and chymotrypsin reacted instantaneously in stoichiometric manner, and the enzyme substrate had no effect on the reaction. The inhibition of plasmin and thrombin was time-dependent and of nonstoichiometric nature, and it was retarded by the presence of enzyme substrate. Even so, kinetic studies failed to show a competition reaction between the substrate and the inhibitor on a single site of the enzyme molecule. It is therefore suggested that the inhibition mechanism might be an enzymatic inactivation of the proteolytic enzymes concerned. This inactivation was irreversible in the sense that no enzymatic activity could be recovered after acid dissociation of the enzyme-inhibitor complex. It was shown by differential titration that the inhibitory activity of the inhibitor towards all four enzymes is due to a single molecular entity; this conclusion is also suggested by the high purity of the inhibitor preparation. On the basis of its stoichiometric combination with trypsin, the cq-proteolytic inhibitor was estimated to have a molecular weight of 47,000 and to comprise about 2% of plasma proteins.

Preparation of Plasminogen by Affinity Chromatography
Background: Plasminogen is one of the compounds derived from human plasma. Activation of plasminogen produces plasmin. Plasmin is able to lyse fibrinogen, fibrin, and some other human plasma proteins. The aim of the present work was to study the separation of human plasminogen by affinity chromatography using gel lysine Sepharose. Materials and Methods: Normal human plasma was used as the...
متن کاملIdentification of molecular forms of plasminogen and plasmin-inhibitor complexes in urokinase-activated human plasma.
Urokinase-activated human plasma was analysed by acetic acid/urea/polyacrylamide-gel electrophoresis. The bands representing plasminogen, the plasmin-alpha 2-plasmin inhibitor and plasmin-alpha 2-macroglobulin complexes were identified by immunoprecipitation with specific antibodies and by comparison with purified components. Plasminogen and the plasmin-inhibitor complexes were isolated from pl...
متن کاملKinetic properties of the primary inhibitor of plasmin from human plasma.
The interaction of human plasmin with the newly discovered alpha2-plasmin inhibitor was investigated. It was found from rate measurements that the reaction involves the rapid formation of a first enzyme-inhibitor complex, followed by the slow irreversible transition to another complex. L-Lysine influences the first step, but not the second.

Human antiplasmin, the fast-acting plasmin inhibitor in plasma, was purified to homogeneity and labeled with 125I. This material, which was indistinguishable from antiplasmin in plasma with respect to several physicochemical and functional properties, was injected intravenously and its turnover measured in control subjects and in patients undergoing thrombolytic therapy. In eight control subjec...

The fraction of human serum designated as C'1 esterase inhibitor is known to inhibit the action of C'1 esterase, a plasma kallikrein, and PF/Dil, an enzyme in plasma enhancing cutaneous vascular permeability. In the present study, C'1 esterase inhibitor has been found to block the actions of plasmin and the C'1r subcomponent of the first component of complement, and to retard the generation of ...
متن کاملAlpha2-plasmin inhibitor and alpha2-macroglobulin-plasmin complexes in plasma. Quantitation by an enzyme-linked differential antibody immunosorbent assay.
An enzyme-linked differential antibody immunosorbent assay has been developed for the quantification of alpha2-plasmin inhibitor-plasmin and alpha2-macroglobulin-plasmin complexes. In this method the inhibitor-plasmin complex is bound to a surface by an inhibitor-specific antibody, and the plasmin bound to the inhibitor is quantified by a second antibody, rabbit antiplasminogen F(ab')2, labeled...
